In vitro study | Docetaxel is a cytotoxic agent, especially for proliferating cells, which, together with its promotion of microtubule bundle formation and induction of sustained mitotic arrest, the ability to subsequently cause mitotic arrest apoptosis or permanent mitotic arrest is correlated. Docetaxel inhibits microtubule dynamical instability as well as microtubule treadmill, resulting in the inability of chromosomes to segregate into the progeny, which in turn causes premature exit from mitosis rather than arrest at the cell cycle stage. Docetaxel inhibits clonal survival of human cancer cells Hs746T (stomach),AGS (stomach),HeLa (cervix),CaSki (cervix),BxPC3 (pancreas),Capan-1 (pancreas), the IC50 values were 1 nM,1 nM,0.3 nM,0.3 nM,0.3 nM and 0.3 nM, respectively. Docetaxels inhibit endothelial cell migration without affecting microtubule gross morphology or inhibiting cell proliferation, although they can have more subtle effects on microtubule dynamics. Docetaxel inhibits HUVEC migration with an observed IC50 of 1 pM. The HUVEC chemotaxis stimulated by two angiogenic factors, Thymidine phosphorylase or VEGF, was inhibited by Docetaxel with an IC 50 of 10 pM and abolished at 1 nM. Docetaxel induces expression of human monocytes without affecting expression of prostaglandin H synthase-2m (PGHS-2) in macrophages from RAW 264.7 mice. |
In vivo study | Docetaxel (33 mg/kg/dose, I. v., 3 injections every 4 days) caused a 19.3-day growth delay in M2OL2 colon cancer xenografts. Docetaxel also showed good antitumor activity in MX-1,SK-MEL-2,LX-1 and OVCAR-3 xenografts. Docetaxel was injected twice a week for 14 days to inhibit the angiogenic response of fibroblast growth factor 2 with an IC 50 of 5.4 mg/kg, and when 10 mg/kg Docetaxel was given, completely blocked angiogenesis in mice. Docetaxel is selective for endothelial cell migration and/or microvascularization because infiltration of inflammatory cells into Matrigel plugs is less sensitive to Docetaxel inhibition. |
